Units per 0.1 mL = units in the vial ÷ (mL of saline × 10). Example: 100 units in 2.5 mL = 100 ÷ 25 = 4 units per 0.1 mL.

How to read the chart

Find the vial size, then the volume of saline you add. The row tells you how many units are in each mL, each 0.1 mL and each 0.05 mL of the reconstituted product. The formula is simple: units per 0.1 mL equals the units in the vial divided by ten times the mL of saline.

The Botox Cosmetic prescribing information reconstitutes a 100-unit vial with 2.5 mL, or a 50-unit vial with 1.25 mL, of preservative-free 0.9% sodium chloride, giving 4 units per 0.1 mL. Those rows are marked on the chart.

Things the chart can't tell you

  • Units aren't interchangeable between brands. A unit of one botulinum toxin product is not the same as a unit of another, so never convert doses between brands with this chart.
  • The diluent, volume, handling, storage and how long reconstituted product can be kept all come from the product's label. Write the use-by on the vial and in the record.
  • Dosing is a clinical decision for the injector and the medical director, not something a dilution chart decides.

Why keep a reconstitution record

Writing down the lot number, volume and time for each vial means every treatment can be traced to a vial if there's a recall, an adverse event or a question about a product's source. It also shows an inspector that reconstituted product isn't kept past its use-by.

Questions

How many units are in 0.1 mL of Botox?

It depends on how much saline was added. A 100-unit vial reconstituted with 2.5 mL, the dilution on the Botox Cosmetic label, gives 4 units per 0.1 mL. With 2 mL it's 5 units per 0.1 mL, and with 1 mL it's 10.

Does this chart work for Dysport, Xeomin or other brands?

The arithmetic works for any vial, but units aren't interchangeable between products and each label has its own reconstitution instructions. Use the chart only for the vial size and volume you actually used, and follow that product's label.
